Black Tea (Camellia sinensis) Steepings Essence is Potensial to Down Regulation The Expression of IGF-1, ERK1/2 and PPARgamma on MAPK (Mitogen Activated Protein of Kinase) Pathways in Adipogenesis Visceral Tissue Wistar Rats with High Fat Diet

Abstract
Obesity prevalence was increase in the various Asian and Pacific Country, Europe, United States, Australian and also in developing country. Change of preadipocyte into mature adipocyte play a part in obesity pathogenesis. Black tea has great amount antioxidant which plays an important role controlling of cells proliferation. Theaflavin in black tea may be able to inhibits change of preadipocyte into mature adipocyte, therefore the prevalence of obesity as inducer various metabolic disease can be prevented early. The goal of this study is to prove the effect of black tea (Camellia sinensis) steepings essence(BTSE) to prevention the raising of IGF-1, ERK1/2 and PPARg expression in wistar rats with high fat diet. The research is conducted by using Complete Divided Randomized Design ( RAL) by in vivo using mouse strain wistar. Experimental animal is divided into positive control group which is given high fat diet without black tea steepings essence and treatments group by high fat diet with black tea steepings essence dose 0,015g, 0,030g and 0,045g/day during 90 day. The expression of IGF-1, ERK1/2 and PPARg is measured by semiquantitative by counting colour intensity at Corell Photo of Paint 11. Intensity colour score of IGF-1, ERK1/2 and PPARg result of imunohistochemistry are analyzed statistically with One way Anova analysis and linear regression with level of significancy p = 0,05. Based on the result of analysis at One way Anova (p<0,05) black tea steepings essence with various dose (0,015g, 0,030g and 0,045g/day) can prevent the raising of IGF-1, ERK1/2 and PPARg expression significantly (p= 0,000) compared to positive control group. At measurement of IGF-1 expression degradation of highest level at dose 0,015g and 0,045g/day. Meanwhile for the expression of ERK1/2 and PPARg degradation of expression both highest at dose 0,045g/day. The greater black tea dose, the lower the expression of IGF-1, ERK1/2 and PPARg will be. Based on the analysis of regression (p<0,05) there are functional relation of two strong direction between ERK1/2 and PPARg (R = 0,798 and R = 0,780). Based on this result study prove that black tea (Camellia sinensis) steepings essence can prevent the increasing of IGF-1, ERK1/2 and PPARg expression in wistar rats with high fat diet.
